Identify experimental units and use paired contrasts

The unit independently assigned to a condition determines the replication available for a treatment claim. A hundred detector scans of one preparation are a hundred technical readings, not a hundred independent biological preparations. They can reveal instrument repeatability while leaving preparation-to-preparation variation almost unmeasured.

Learning outcome: Set treatment effect0 and batch shift4. Compare the paired estimate with the confounded estimate. Then change the treatment effect while holding batch offset fixed. State the number of biological units and name a violation that pairing cannot remove.

Starting recap

Separate mechanism, measurement, material balance and model validity. Preserve raw evidence and the assumptions needed for each inference.

All lessons are open directly. Your saved experiments remain when you leave and return.

Try

Baselines8,10,12, a treatment effect3 and a shared+2 shift in preparation3 give paired controls8,10,14 and treated11,13,17. Each paired difference is3. A separate confounded design with only treatment batches shifted gives an apparent contrast3+2=5.

Open the experiment

Predict

Will a common shift in one paired preparation change its treatment-minus-control difference? Would shifting only the treated batch cancel?

Compare treatment contrasts within biological blocks

Worked teaching model · synthetic values · Three independent fictional biological preparations have baseline responses 8,10,12. Each preparation is split into paired control/treatment aliquots; offset affects both aliquots of preparation3. The alternative confounded comparison uses unshifted controls and shifted treatment batches. Splits and technical repeats are not additional independent preparations.

Reset keeps your prediction, captures and practice history.

Within-preparation contrasts cancel a common additive batch shift; confounded assignment does not.

Pairing is useful because the comparison shares relevant nuisance factors. It does not remove treatment-by-preparation interactions, spillover or measurement drift that differs between aliquots. Three paired preparations, not six aliquots or repeated scans, are the independent biological units in this stipulated design.

Understand

1. Orient: Identify experimental units and use paired contrasts

The unit independently assigned to a condition determines the replication available for a treatment claim. A hundred detector scans of one preparation are a hundred technical readings, not a hundred independent biological preparations. They can reveal instrument repeatability while leaving preparation-to-preparation variation almost unmeasured.

2. Explain the mechanism and assumptions

Blocking groups comparable units so that a treatment comparison is made within each group. Our three independent fictional preparations are each divided into control and treatment aliquots. The paired difference within each preparation removes a shared additive preparation shift. The three differences, not six aliquots or repeated scans, are the three independent biological contrasts under this design.

Random assignment within a block helps prevent a handling position from always receiving one treatment. Blinding reduces expectation-driven measurement or classification. Neither guarantees that every causal alternative has disappeared: spillover between aliquots, treatment-specific drift and interactions with preparation state still require attention. Preserve the allocation rule and any missing pairs before analysis.

An unpaired comparison can become confounded if all treated preparations come from a shifted batch and all controls from another batch. Subtracting their means then combines treatment and batch effects. More observations of the same confounded allocation can make a biased estimate look precise. The model contrasts this design with pairing; it does not calculate a confidence interval or claim that three preparations are sufficient for every biological question.

3. Work a quantitative example

Baselines8,10,12, a treatment effect3 and a shared+2 shift in preparation3 give paired controls8,10,14 and treated11,13,17. Each paired difference is3. A separate confounded design with only treatment batches shifted gives an apparent contrast3+2=5.

4. Predict and investigate

Predict first: Will a common shift in one paired preparation change its treatment-minus-control difference? Would shifting only the treated batch cancel?

Set treatment effect0 and batch shift4. Compare the paired estimate with the confounded estimate. Then change the treatment effect while holding batch offset fixed. State the number of biological units and name a violation that pairing cannot remove.

Retain the initial prediction, model settings, raw displayed quantities and your interpretation in the lesson notebook. All generated observations are explicitly synthetic; no wet-laboratory procedure is required.

5. Transfer to a changed case

Try a changed case before opening the response

Four preparations each provide a paired control and treatment aliquot, with three detector scans per aliquot. How many independent biological paired contrasts are available? What do the additional scans contribute?

Write your calculation, units and model boundary in the retained lesson notebook first. This worked response is public practice, not independent assessment.

Compare with a worked response

There are four independent biological paired contrasts, not eight or24. Repeated scans assess technical repeatability; aliquots share preparation-level influences. The inference also depends on assignment, independence between preparations and a justified biological sampling frame.

6. Reflect and retain a defensible claim

Explain which result would weaken your favored mechanism. Distinguish the biological unit from repeated readings, included uncertainty from omitted bias, and a model calculation from a performed physical investigation. State one additional control or independent observation needed before generalizing beyond this case.

Save a science backup or import one

Science work is stored locally in this browser, separately from mathematics and AI. It does not sync automatically. Use this science-only backup; this checkout's older all-subject export does not include science.

A legacy file is available only when the complete snapshot fits the 36 MB limit. For larger snapshots, prepare a complete file set below; never discard notebooks to make a smaller file.

Matching imported notebook drafts replace their current drafts. Omitted lessons and known first attempts remain. Export your current work first if you want to retain both drafts.

Complete file sets and course backups

A file set can hold more work than one science file. Keep its manifest and every part together. Each file is limited to 36 MB. This format needs this version of LearnAI or a compatible newer version; older versions can use only the legacy single-file backup above.

The set includes every saved science course and global continuation at this moment. Later edits need a new export. A conflicting tab exports its own visible work, which may differ from saved work in another tab.

0 file(s) selected. File order and filenames do not determine identity. Missing, changed or conflicting parts block the entire restore; no good part is applied by itself.
