Separate receptor binding from a cellular response

A signal reaches a cell only through a chain of physical and molecular events. Local paracrine signals, circulating endocrine signals, self-directed autocrine signals and direct cell contacts differ in delivery. None guarantees a response in every nearby cell: receptor expression and downstream machinery determine which cells can interpret a signal.

Learning outcome: Compare receptor amounts 20 and 80 at ligand 2, then compare ligand 2 and 6 at fixed amount. Retain fraction and amount separately. Propose a downstream readout and a control that could reveal equal binding but different signal interpretation.

Starting recap

Separate receptor binding, downstream response and checkpoint passage. Declare DNA-counting boundaries, population age assumptions and the evidence needed to distinguish mechanisms.

All lessons are open directly. Your saved experiments remain when you leave and return.

Try

With Kd=2 nmol/L and free ligand 2 nmol/L, occupancy is 1/2. A cell with 60 receptor units has 30 bound units; a reference with 20 has 10. Both have the same affinity and occupied fraction. At free ligand 6, the fraction rises to 3/4.

Does doubling receptor amount change the ligand concentration required for half occupancy under the excess-ligand assumption?

Worked teaching model · synthetic values · Independent equilibrium binding sites with Kd=2 nmol/L and ligand held in excess. Bound fraction=L/(Kd+L). Binding is not automatically a proportional cellular response; downstream context is not modeled.

Receptor abundance changes bound amount without changing affinity in this model.

Half occupancy occurs at free ligand Kd, regardless of total receptor amount under the excess-ligand assumption. Different cellular responses still require downstream context.

Understand

1. Frame the biological question

2. Explain mechanisms and assumptions

A water-soluble ligand commonly acts through a cell-surface receptor because crossing the lipid interior is unfavorable. Some small hydrophobic signals can cross membranes and bind intracellular receptors that regulate gene expression. Ligand-gated channels instead change ion conductance; their effect depends on ion gradients as well as channel opening. Receptor binding is therefore a starting condition, not one universal response mechanism.

For independent binding sites at equilibrium, the supplied fraction occupied is L/(Kd+L), with free ligand L and dissociation constant Kd. At L=Kd half the sites are occupied. We hold free ligand in excess so receptor binding does not appreciably deplete it. If that assumption fails, a mass balance must replace the fixed-free-ligand simplification.

Total bound amount equals occupied fraction times receptor abundance. Increasing abundance can increase this amount without changing affinity. Conversely, two cells can have equal receptor occupancy but different outputs because one lacks a relay, expresses an inhibitor or regulates different target genes. A response curve is not automatically a binding curve, and its half-response concentration need not equal Kd.

3. Work a quantitative example

4. Predict and investigate

Predict first: Does doubling receptor amount change the ligand concentration required for half occupancy under the excess-ligand assumption?

Compare receptor amounts 20 and 80 at ligand 2, then compare ligand 2 and 6 at fixed amount. Retain fraction and amount separately. Propose a downstream readout and a control that could reveal equal binding but different signal interpretation.

Retain the initial prediction, settings, raw quantities and interpretation in the lesson notebook. These are supplied or calculated teaching records, not observations from a performed biological experiment.

5. Apply the reasoning to a changed case

Apply the mechanism to a changed case

An independent-site receptor has Kd=3 nmol/L, free ligand 9 nmol/L and total amount 80 units. Find occupied fraction and bound amount. Does this uniquely determine transcription?

Retain your calculation, units and assumptions in the lesson notebook before opening this public practice response.

Compare a worked response

Fraction is 9/(3+9)=0.75 and bound amount is 60 units. Transcription is not determined without receptor mechanism, downstream components and target-gene context.

6. Reflect and retain evidence

Which observation would distinguish the proposed mechanism from a measurement artifact or an omitted process? Define the biological unit, controls, counting boundary and uncertainty. A correct calculation or submitted plan does not certify supervised practical skill or independently demonstrated mastery.

Save a science backup or import one

Science work is stored locally in this browser, separately from mathematics and AI. It does not sync automatically. Use this science-only backup; this checkout's older all-subject export does not include science.

A legacy file is available only when the complete snapshot fits the 36 MB limit. For larger snapshots, prepare a complete file set below; never discard notebooks to make a smaller file.

Matching imported notebook drafts replace their current drafts. Omitted lessons and known first attempts remain. Export your current work first if you want to retain both drafts.

Complete file sets and course backups

A file set can hold more work than one science file. Keep its manifest and every part together. Each file is limited to 36 MB. This format needs this version of LearnAI or a compatible newer version; older versions can use only the legacy single-file backup above.

The set includes every saved science course and global continuation at this moment. Later edits need a new export. A conflicting tab exports its own visible work, which may differ from saved work in another tab.

0 file(s) selected. File order and filenames do not determine identity. Missing, changed or conflicting parts block the entire restore; no good part is applied by itself.
