Interpret organelle enrichment using independent marker controls

Eukaryotic compartments organize processes while remaining connected by transport and signaling. The nucleus encloses much of the genetic material; ribosomes synthesize proteins; endoplasmic reticulum and Golgi participate in processing and traffic; mitochondria and chloroplasts have distinct energy-transforming roles. A compartment name is a hypothesis about location and function, not a substitute for evidence.

Learning outcome: Hold true enrichment4 and vary contamination0,20 and50%. Record both marker signals and target enrichment. Explain why a target-enriched fraction is not itself proof of direct target–marker binding or of complete organelle recovery.

With20% cytosolic contribution and true organelle target enrichment4-fold, target signal is0.8×80+0.2×20=68units per protein. Observed enrichment over cytosol is68/20=3.4-fold. Marker readings80 and20 describe the stipulated mixture; they do not reveal total organelle yield.

Will contamination always make the observed target enrichment equal to its pure-organelle enrichment?

Interpret compartment enrichment with contamination controls

Worked teaching model · synthetic values · Original mixture model per equal total protein: organelle marker100units in pure organelle and0 in cytosol; cytosol marker conversely100. Target is20units in cytosol and20×enrichment in organelle. No separation protocol, yield, activity, cell purity or absolute organelle abundance is inferred.

Reset keeps your prediction, captures and practice history.

Contamination dilutes apparent enrichment; marker controls describe the mixture, not absolute recovery.

Equal-total-protein normalization permits the stipulated mixture calculation. Co-fractionation can support localization but does not prove direct interaction, exclusive location or function. Orthogonal localization evidence and marker specificity matter.

Understand

1. Orient: Interpret organelle enrichment using independent marker controls

2. Explain the mechanism and boundary

Fraction enrichment compares a target signal in a selected material fraction with an appropriate reference. Equal-total-protein normalization helps compare concentrations of detected material, but it does not give total recovery or the number of organelles. A target can appear enriched because of its distribution, contamination, unequal recovery or measurement differences.

The original mixture model uses two markers: an organelle marker present only in the stipulated organelle material and a cytosol marker present only in cytosol. Their known responses estimate contamination under those assumptions. The target itself is20units per protein in cytosol and a selected multiple of that in pure organelle material.

Co-fractionation supports shared distribution only within the method’s resolution. It does not prove direct molecular binding, exclusive localization or biological function. Orthogonal imaging, a second independent marker and a check of marker specificity can challenge the interpretation. No physical isolation or centrifugation procedure is requested here; the task is an inference audit on supplied mixtures.

3. Work a quantitative example

4. Predict and investigate

Predict first: Will contamination always make the observed target enrichment equal to its pure-organelle enrichment?

Hold true enrichment4 and vary contamination0,20 and50%. Record both marker signals and target enrichment. Explain why a target-enriched fraction is not itself proof of direct target–marker binding or of complete organelle recovery.

Retain your initial prediction, control settings and all relevant displayed evidence in the lesson notebook. The schematic and calculated records are original synthetic teaching material, not measurements of a physical specimen.

5. Transfer and explain

Apply the mechanism to a changed case

A mixture contains25% cytosolic protein. Target signal is20units per protein in cytosol and100 in pure organelle material. Predict the mixture signal and apparent enrichment over cytosol.

Retain your calculation, units and assumptions in the lesson notebook before opening this public practice response.

Compare a worked response

Mixture signal=0.75×100+0.25×20=80units per protein; apparent enrichment=80/20=4-fold, below the pure-organelle5-fold enrichment. The result depends on linear signal mixing and the normalization assumptions.

6. Reflect and retain evidence

Explain the link from cellular structure or membrane process to the modeled observation. State a competing explanation, an omitted variable and one independent comparison that could challenge your conclusion. Do not turn a simulated observation into a claim of laboratory participation or independently demonstrated practical skill.

What does target co-enrichment with an organelle marker establish by itself?

Public practice checks a specific claim. It does not establish independent mastery.

A mixture contains25% cytosolic protein. Target signal is20units per protein in cytosol and100 in pure organelle material. Predict the mixture signal and apparent enrichment over cytosol.

Completion requires a saved prediction, at least one capture and a reflection. Presence checks do not grade your reasoning.

Skipping retains experiments and awards no mastery.

Save a science backup or import one

Science work is stored locally in this browser, separately from mathematics and AI. It does not sync automatically. Use this science-only backup; this checkout's older all-subject export does not include science.

A legacy file is available only when the complete snapshot fits the 36 MB limit. For larger snapshots, prepare a complete file set below; never discard notebooks to make a smaller file.

Matching imported notebook drafts replace their current drafts. Omitted lessons and known first attempts remain. Export your current work first if you want to retain both drafts.

Complete file sets and course backups

A file set can hold more work than one science file. Keep its manifest and every part together. Each file is limited to 36 MB. This format needs this version of LearnAI or a compatible newer version; older versions can use only the legacy single-file backup above.

The set includes every saved science course and global continuation at this moment. Later edits need a new export. A conflicting tab exports its own visible work, which may differ from saved work in another tab.

0 file(s) selected. File order and filenames do not determine identity. Missing, changed or conflicting parts block the entire restore; no good part is applied by itself.

Science storage: loading. No account or remote computation is required.
