Distinguish passage through a checkpoint from successful repair

Cell-cycle progression depends on regulated protein activity, not an automatic clock alone. Cyclins and cyclin-dependent kinases coordinate transitions, while inhibitors and checkpoint pathways can restrain progression when replication, DNA integrity or spindle attachment is inappropriate. Cyclin abundance, kinase modification and protein degradation contribute different kinds of control.

Learning outcome: Compare intact and bypassed settings at damage counts 20 and 80. Keep undamaged passage, damaged passage and arrest separate. Specify an independent readout that would be required for a repair claim, and one for a survival claim.

Starting recap

Separate receptor binding, downstream response and checkpoint passage. Declare DNA-counting boundaries, population age assumptions and the evidence needed to distinguish mechanisms.

All lessons are open directly. Your saved experiments remain when you leave and return.

Try

With 40 of 100 cells labeled damaged, the intact checkpoint permits 60 undamaged cells to proceed and arrests 40. Bypass permits all 100 to proceed, including 40 damaged cells. The increased passage count provides no evidence that those 40 were repaired.

Open the experiment

Predict

If more cells proceed after bypass, which missing measurement prevents concluding that repair improved?

Separate checkpoint arrest from viability and repair

Worked teaching model · synthetic values · Synthetic 100-cell cohort: intact checkpoint arrests all assigned damaged cells, while bypass permits all cells to proceed. No repair, death, proliferation rate or medical outcome is modeled. This all-or-none rule is a teaching contrast, not real checkpoint sensitivity.

Reset keeps your prediction, captures and practice history.

More cells passing a checkpoint does not demonstrate more repair or better viability.

Cyclin–CDK activity promotes transitions; checkpoint pathways can inhibit progression until conditions are acceptable. This cohort has no readout of repair, apoptosis or eventual offspring survival. Do not infer any treatment recommendation.

Understand

1. Frame the biological question

2. Explain mechanisms and assumptions

A G1 decision integrates growth conditions and damage signals before replication; G2 controls entry into mitosis after replication; spindle-attachment surveillance helps prevent premature sister separation. These controls are not identical sensors. A DNA-damage response may allow repair, prolong arrest or contribute to apoptosis depending on context; no single outcome is guaranteed by the word checkpoint.

The synthetic cohort contains 100 cells, a chosen number of which are labeled damaged. An intact all-or-none checkpoint arrests those cells; an artificial bypass lets them proceed. The model deliberately has no repair or survival variable. It can test an inference about passage but cannot establish that bypass restores genome integrity or produces healthy descendants.

Unregulated growth can follow disruption of several interacting controls, but a simple simulated checkpoint contrast is not a diagnosis or treatment model. Compare progression, damage, repair and survival with separate validated readouts in a conceptual design. No manipulation of actual cells, drugs or organisms is requested, and written design work does not substitute for approved supervised laboratory practice.

3. Work a quantitative example

With 40 of 100 cells labeled damaged, the intact checkpoint permits 60 undamaged cells to proceed and arrests 40. Bypass permits all 100 to proceed, including 40 damaged cells. The increased passage count provides no evidence that those 40 were repaired.

4. Predict and investigate

Predict first: If more cells proceed after bypass, which missing measurement prevents concluding that repair improved?

Compare intact and bypassed settings at damage counts 20 and 80. Keep undamaged passage, damaged passage and arrest separate. Specify an independent readout that would be required for a repair claim, and one for a survival claim.

Retain the initial prediction, settings, raw quantities and interpretation in the lesson notebook. These are supplied or calculated teaching records, not observations from a performed biological experiment.

5. Apply the reasoning to a changed case

Apply the mechanism to a changed case

In a supplied cohort of 200 cells, 30% carry a damage label. Under the same all-or-none rule, compare intact and bypassed passage and state what remains unknown.

Retain your calculation, units and assumptions in the lesson notebook before opening this public practice response.

Compare a worked response

There are 60 labeled cells. Intact control permits 140 to proceed and arrests 60; bypass permits 200, including the 60 labeled cells. Repair, eventual survival and clinical meaning remain unmeasured.

6. Reflect and retain evidence

Which observation would distinguish the proposed mechanism from a measurement artifact or an omitted process? Define the biological unit, controls, counting boundary and uncertainty. A correct calculation or submitted plan does not certify supervised practical skill or independently demonstrated mastery.

Save a science backup or import one

Science work is stored locally in this browser, separately from mathematics and AI. It does not sync automatically. Use this science-only backup; this checkout's older all-subject export does not include science.

A legacy file is available only when the complete snapshot fits the 36 MB limit. For larger snapshots, prepare a complete file set below; never discard notebooks to make a smaller file.

Matching imported notebook drafts replace their current drafts. Omitted lessons and known first attempts remain. Export your current work first if you want to retain both drafts.

Complete file sets and course backups

A file set can hold more work than one science file. Keep its manifest and every part together. Each file is limited to 36 MB. This format needs this version of LearnAI or a compatible newer version; older versions can use only the legacy single-file backup above.

The set includes every saved science course and global continuation at this moment. Later edits need a new export. A conflicting tab exports its own visible work, which may differ from saved work in another tab.

0 file(s) selected. File order and filenames do not determine identity. Missing, changed or conflicting parts block the entire restore; no good part is applied by itself.

Science storage: loading. No account or remote computation is required.
