Trace antiparallel replication and repair limits

DNA replication is semiconservative: each daughter duplex contains one parental strand and one newly synthesized strand.

Learning outcome: Write the complementary strand to 5′-ACGTTA-3′ first aligned beneath it, then in 5′→3′ notation.

Starting recap

DNA replication is semiconservative: each daughter duplex contains one parental strand and one newly synthesized strand. Complementary pairing allows each strand to guide copying, but polymerase extends a new strand in the 5′ to 3′ direction by adding to its 3′ end.

Predict

Before opening the changed condition, predict which measurement would discriminate the explanation in trace antiparallel replication and repair limits.

Experiment

Select “Trace antiparallel replication and repair limits” in the case control. Compare both evidence conditions. Record the quantities, calculate the worked comparison and justify your claim with a stated alternative. The supplied records are synthetic practice, not a conducted experiment.

The counts omit degradation and do not measure synthesis rate.

Compare with the conditional interpretation

Two daughter duplexes each contain one old and one new strand.

For template 3′-TAC GGA-5′, complementary new DNA is 5′-ATG CCT-3′. A response written 3′-ATG CCT-5′ has the right letters but the wrong orientation. In generation2, original-strand fraction is 2/8=1/4.

Understand

Mechanism before a label

DNA replication is semiconservative: each daughter duplex contains one parental strand and one newly synthesized strand. Complementary pairing allows each strand to guide copying, but polymerase extends a new strand in the 5′ to 3′ direction by adding to its 3′ end.

At a replication fork, the two templates have opposite orientations. Leading-strand synthesis can proceed continuously toward the advancing fork, while lagging-strand synthesis uses discontinuous fragments that are later joined. Helicase unwinds, primase supplies starting primers and ligase joins appropriate discontinuities; these roles should not be collapsed into one generic enzyme.

Proofreading and repair reduce copying errors but do not guarantee zero mutation. A mismatch is not necessarily a permanently inherited change: repair may remove it. Track both sequence orientation and which generation contains a stable change before making an inheritance claim.

Separate opening from torsional relief

Helicase separates the template strands at the fork. Unwinding can create torsional strain ahead of it; topoisomerase relieves that strain by transiently changing DNA strand connectivity and resealing it. This is not the same role as helicase, polymerase extension or ligase sealing remaining backbone discontinuities. DNA polymerase extends a pre-existing primed end; RNA primers supply starting points in the cellular replication model. OpenStax replication machinery, checked 31 October 2026.

Changed case: A supplied cartoon contains a primed template and functioning polymerase but progressively tighter twisting ahead of its advancing fork. Which missing function is most directly implicated?

Check the replication role

Relief of torsional strain implicates topoisomerase function. Adding another primer does not directly remove that twisting; ligase sealing is a different backbone task. This identifies a role in a stipulated cartoon, not a diagnosis from one real biological observation.

Trace a short coding sequence

This original toy sequence is not a real gene, gene-design tool or wet-lab instruction. The supplied DNA is the coding strand, 5′→3′. This exercise deliberately starts its reading frame at the first base, uses the standard genetic code, and stops at the first in-frame stop. It does not infer initiation, splice RNA, process introns or model alternative genetic codes.

Predict first: change GCT to GCC (both alanine, A), then change it to TAA (stop). Finally insert one C after ATG and compare downstream triplets. Preserve your first prediction in the lesson notebook before editing. This scratch input is not saved or graded.

Worked reasoning

For template 3′-TAC GGA-5′, complementary new DNA is 5′-ATG CCT-3′. A response written 3′-ATG CCT-5′ has the right letters but the wrong orientation. In generation2, original-strand fraction is 2/8=1/4.

Two daughter duplexes each contain one old and one new strand.

Apply to a new case

Write the complementary strand to 5′-ACGTTA-3′ first aligned beneath it, then in 5′→3′ notation.

Compare your reasoning with the worked response

The aligned complement is 3′-TGCAAT-5′. Reversing the written order gives 5′-TAACGT-3′. Complementing and reversing are separate operations.

This public response supplies feedback, not a score for independent reasoning. Record which part of your prediction changed and why.

Evidence and investigation boundary

The counts omit degradation and do not measure synthesis rate. Density-label predictions require label incorporation and separation assumptions not supplied by counts alone.

For a supervised investigation, predeclare the question, biological unit of replication, treatment/comparison assignment, quantities and units, exclusion rules, graph and uncertainty method. Keep the raw records and the first prediction separate from later revisions. A simulated case does not supply laboratory hours or establish AP authorization. No physical manipulation is required by this lesson.

Save a science backup or import one

Science work is stored locally in this browser, separately from mathematics and AI. It does not sync automatically. Use this science-only backup; this checkout's older all-subject export does not include science.

A legacy file is available only when the complete snapshot fits the 36 MB limit. For larger snapshots, prepare a complete file set below; never discard notebooks to make a smaller file.

Matching imported notebook drafts replace their current drafts. Omitted lessons and known first attempts remain. Export your current work first if you want to retain both drafts.

Complete file sets and course backups

A file set can hold more work than one science file. Keep its manifest and every part together. Each file is limited to 36 MB. This format needs this version of LearnAI or a compatible newer version; older versions can use only the legacy single-file backup above.

The set includes every saved science course and global continuation at this moment. Later edits need a new export. A conflicting tab exports its own visible work, which may differ from saved work in another tab.

0 file(s) selected. File order and filenames do not determine identity. Missing, changed or conflicting parts block the entire restore; no good part is applied by itself.
